What controls matter most when reading the specificity claims?

For a result like 'cleaves the intended site', the controls do the heavy lifting: scrambled/ mismatched guides, no-guide conditions, and off-target site panels. In vitro specificity also doesn't guarantee in-cell specificity. What off-target controls would you consider essential before believing a programmability claim?
